Western blot

Sample

WT and Pcgf1−/− ES cells. Cells were harvested and lysed in hypotonic lysis buffer (10 mM Tris–Cl pH 8.0, 1 mM KCl, 1.5 mM MgCl2 and 0.5mM β-mercaptoethanol, 10 mg/ml PMSF, Protease Inhibitor Mix (Sigma)).

Blocking agent

5% (w/v) non-fat milk for one hour at room temperature.

Primary incubation

1:1,000 dilution overnight at 4 °C.

Secondary incubation

1:5,000 dilution horseradish peroxidase-conjugated goat anti-mouse lgG-HRP (sc-2005, Santa Cruz Biotechnology).

Detection

ECL blot detection system.
