Sample

WT and PARP1 KO OVCAR8 cells.

Blocking agent

WB: LiCor Odyssey blocking buffer for 1 h.

Primary incubation

ICC: 1:1,000 dilution for 1 h.

WB: 1:1,000 dilution in Odyssey blocking buffer for 1h incubation at 37C.

Secondary incubation

ICC: goat anti-rabbit IgG Alexa Fluor Plus 488 (cat# A327731) for 1 h.

WB: 1:10,000 dilution secondary antibodies (Thermo Fisher, Philadelphia, PA, USA) in Odyssey blocking buffer and incubated at 37C for 1 h.

Detection

ICC: Images were taken on a Zeiss Observer Microscope and fluorescent intensity was quantified using Zeiss Zen software (Zeiss, Netherlands).

WB: LiCor imager.
