OGG1/2 antibody | knockout validation | Santa Cruz sc-376935

This is a knockout-validated antibody summary, based on the publication "CHD4 Has Oncogenic Functions in Initiating and Maintaining Epigenetic Suppression of Multiple Tumor Suppressor Genes", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1 κ (kappa light chain)

Company: Santa Cruz

Antibody: OGG1/2

Catalog number: sc-376935

Summary: Mouse monoclonal IgG1 κ (kappa light chain) against an epitope mapping between amino acids 13-49 near the N-terminus of human OGG1/2. Recommended for detection of OGG1 and OGG2 of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence and ELISA.

Validation Method

Western blot

Sample

WT, OGG1-KO and OGG1 recovered SW480 cells.

Blocking agent

5% milk in TBST (120 mM Tris–HCl (pH 7.4), 150 mM NaCl, and 0.05% Tween 20) for 1 hr at room temperature.

Primary incubation

Overnight at 4 °C.

Secondary incubation

HRP-conjugated secondary antibody.

Detection

Dura SuperSignal Substrate (Pierce, USA).

Figure

After SW480 OGG1 KO cells were transfected with pCMV-Taq or pCMV-OGG1 for 48 hr, the cells were untreated or treated with 2mMH2O2 for 30 min. Whole-cell extracts and the tight chromatin fractions were analyzed by immunoblotting with the indicated antibodies. Please see Figure 5c in the article [1].
