This is a knockout-validated antibody summary, based on the publication "NOVA2-mediated RNA regulation is required for axonal pathfinding during development", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 2. Immunohistochemistry of coronal brain sections of E16.5 wild-type, Nova1-/-, and Nova2-/-, stained for NOVA1 (green), NOVA2 (red), and DAPI (blue). Scale bar; 500 μm. From [1].
Sample

WB: Extracts of mouse cortex from age-matched P0 wild-type, Nova2-/-, and Nova1-/- mice. IHC: E16.5, E18.5 mice brains were fixed with 4% paraformaldehyde (PFA)/PBS at 4 C overnight, and embedded in OCT compound for cryosection (80 μm, CM3050S, LEICA).

Blocking agent

IHC: 1.5% normal donkey serum (NDS)/PBS at RT.

Primary incubation

IHC: overnight at 4 C.

Secondary incubation

IHC: 1:1,000 dilution Alexa 488, 555, or 647 conjugated donkey secondary antibodies in 1.5% NDS/PBS.

Detection

IHC: Images of immunostained specimens were collected by BZ-X700 (KEYENCE), fluorescence microscopes Axioplan 2 (Zeiss), or an inverted TCS SP8 laser scanning confocal microscope (LEICA).
