This is a knockout-examined antibody summary, based on the publication "Physical Interactions and Functional Relationships of Neuroligin 2 and Midbrain Serotonin Transporters", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Western blots of NLGN2 protein in NLGN2 null mice (KO), NLGN2 heterozygous (HET) and their WT littermates in midbrain. GAPDH is used as loading control. From figure 1 [1]. The band at a higher molecular weight in NLGN2 null lane is thought to be NLGN3.
Antibody information

Antibody: NLGN2

Catalog number: 129211

Summary: Mouse monoclonal IgG1 against neuroligin 1/2/3/4, raised against recombinant mouse neuroligin 3 (cytoplasmic tail, aa 608 - 722). Reacts with rat and mouse. In western blot recognizes neuroligin 2 and 3.

Immunocytochemistry, stains predominantly neuroligin 2 in cultured hippocampal neurons. Detects transiently expressed GFP fusion proteins comprising the cytoplasmic tails of neuroligins 1-4.

Company: Synaptic Systems

Method

Western Blot

Sample

Freshly dissected brain tissues, homogenization in RIPA buffer (50 mM Tris, pH 7.4, 150 mM NaCl, 1 mM EDTA, 1% TRITON X-100, 1% sodium deoxycholate, 0.1% SDS) containing protease inhibitors (P8340, 1:100; Sigma, St. Louis, MO, USA).

Primary incubation

1:1,000

Detection

chemiluminescence (Western Lightning ECL Pro, Perkin Elmer) and X-ray film (Biomax MS, Kodak, 8294985) exposure.
