Sample

Whole brain lysate of NLG1 KO and WT mice. Tissues were lysed for 45 min in ice-cold cell lysis buffer containing 20 mM Tris, pH 7.5, 150 mM NaCl, 1 mM EDTA, 1 mM EGTA, 1% Triton X-100, 2.5 mM sodium pyrophosphate, 1 mM β-glycerophosphate, 1 mM Na3VO4, 20 mM NaF, and 1% protease inhibitor cocktail and phosphatase inhibitor (Roche).

Blocking agent

5% dry milk TBST (20 mM Tris base, 9% NaCl, 1% Tween-20, pH 7.6).

Primary incubation

1:2,000 dilution in TBST overnight at 4°C.

Secondary incubation

1:2,000 dilution goat anti-rabbit (A00098, Genscript).

Detection

Enhanced chemiluminescence (Thermo Fisher Scientific).
