Immunohistochemistry

Sample

IHC: Mouse kidneys from control, Hoxb7:Cretg/+ (Nf2UB-/-) and Six2:CreTGC/+ (Nf2CM-/-) conditional deletion of Nf2. Tissues were fixed in 4% paraformaldehyde and embedded in paraffin for sectioning.

Blocking agent

IHC: 3% BSA, 10% goat serum, 0.1% Tween20 in PBS at room temperature.

Primary incubation

WB: 1:2,000 dilution.

IHC: 1:300 dilution in 3% BSA, 3% goat serum and 0.1% Tween20 in PBS.

Secondary incubation

IHC: conjugated secondary antibodies (Jackson Laboratories).

Detection

IHC: Nikon C1 plus Digital Eclipse confocal microscope.

Notes

Western is not the best example of validation, since protein levels in KO are comparable with WT due to deletion in only one type of cells.
