This is a knockout-validated antibody summary, based on the publication "Polysialic acid as an antigen for monoclonal antibody HIgM12 to treat multiple sclerosis and other neurodegenerative disorders", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Western blot showing binding of the PSA-NCAM antibody (clone 2-2B) and the NCAM antibody (rabbit, Millipore, AB5032) and (rat, Millipore, MAB310) and β-actin (loading control) in brain lysates from heterozygous mice and homozygous NCAM knockout mice lacking all three major splice variants resulting in NCAM180, NCAM140 and NCAM120 expression. Antibody information

Rabbit polyclonal IgG

Company: Millipore

Antibody: NCAM

Catalog number: AB5032

Summary: Rabbit polyclonal IgG against highly purified chicken NCAM. Reacts with chicken, human, mouse and rat. Suitable for western blot, immunofluorescence, immunohistochemistry, function blocking and ELISA.

Validation Method

Western blot

Sample

Brain homogenates from NCAM total KO mice and heterozygous littermate controls.

Primary incubation

1:3,000 dilution.

Secondary incubation

1:5,000 dilution horseradish peroxidase (HRP) conjugated goat anti rabbit IgG (H+L), (Millipore, AP132P).

Notes

Unlike in brain, in liver and other tissues the antibody might not be specific, possibly due to other proteins or “leaky” KO mice.
