Control and MlckSMKO internal anal sphincter (IAS) smooth-muscle cells. IAS smooth muscle strips were homogenated in 120 μl lysis buffer (containing 20 mM Tris base, 137 mM NaCl, 2 mM EDTA, 1% NP-40, 10% glycerol, protease inhibitor cocktail (Roche), pH=8).

Primary incubation

1:10,000 dilution.

Detection

Super Signal West Dura substrate (PIERCE) and MaxiSignal Western Solution (SUDGEN) were used.
