Western blot | Immunocytochemistry

HCT116p53+/+ and HCT116p53−/− cells. WB: Total protein was extracted using RIPA buffer supplemented with protease and phosphatase inhibitors. IC: in 4% paraformaldehyde for 20 min, permeabilized with 0.1% Triton X-100 for 10 min.

WB: 5% dry milk in tris-buffered saline/0.1% Tween-20.

WB: overnight at 4°C.

IC: 1:100 dilution for 1 h at 37°C .

WB: 1:5,000 dilution horseradish peroxidase-conjugated secondary antibody for 2 h.

IC: 1:200 dilution mouse anti-rabbit IgG conjugated with FITC (Invitrogen) for 30 min at 37°C.

IC: LSM510 confocal microscope (Carl Zeiss, Gottingen, Germany).
