Sample

Intestinal epithelium from control and Msi1/2flx/flx::Villin-CreERT2 (Msi-DKO) mice. Tissues were fixed in 4% PFA overnight at 4°C, washed in PBS, and moved to 70% ethanol before paraffin embedding and sectioning.

Primary incubation

1:200 dilution.

Secondary incubation

Cy2-, Cy3-, and Cy5-conjugated secondary antibodies (Jackson ImmunoResearch Laboratories, Inc.)

Detection

Leica Biosystems SP5 (LAS AF software) and SP8 confocal microscopes (LAS X software) were used for fluorescence microscopy equipped with 10× dry, 20× dry, 40× water, and 63× oil objectives.

Musashi-2 antibody (1:200, NBP1-42029; Novus Biologicals) is also KO validated in the same Figure.
