This is a knockout-validated antibody summary, based on the publication "FOXO1 couples metabolic activity and growth state in the vascular endothelium", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody

MYC, also called MRTL; MYCC; bHLHe39; c-Myc; avian myelocytomatosis viral oncogene homolog; class E basic helix-loop-helix protein 39; myc-related translation/localization regulatory factor; proto-oncogene c-Myc; transcription factor p64; v-myc myelocytomatosis viral oncogene homolog

Catalog number

Catalog number is 06-340.

Summary

Rabbit polyclonal antibody that detects Myc protein from human, mouse, avian and xenopus. It has been validated in ChIP, immunocytochemistry, IP and Western blot.

Company

Millipore, now part of MilliporeSigma.

Validation Method

IHC, immunohistochemistry.

Sample

whole mouse eyes were fixed in 4% paraformaldehyde (PFA) on ice for 1 h. Eyes were washed in PBS before the retinas were dissected and partially cut into four leaflets.

Blocking agent

blocking/permeabilization in 2% goat serum (Vector Laboratories), 1% BSA and 0.5% Triton X-100 (in PBS) for 1 h at room temperature.

Primary incubation

incubated at 4 °C overnight in incubation buffer containing 1% goat serum, 0.5% BSA and 0.25% Triton X-100 (in PBS) and the primary antibody 1:100.

Secondary incubation

After four washes with 0.1% Triton X-100 in PBS (PBST), retinas were incubated with Alexa-Fluor 488-, Alexa-Fluor 555- or Alexa-Fluor 647-conjugated secondary antibodies (Invitrogen, 1:400) for 2 h at room temperature.

Detection

flat-mounted with Vectashield (Vector Laboratories) and examined by confocal laser microscopy (Leica TCS SP5 or SP8).

Notes

No clear image as KO is only in endothelial cells.

Figure

See Fig 4 in the [1]. Immunofluorescence of Myc protein (in red) in control and Myc-KO endothelial cells of mouse retinas. VECAD (in grey) and PECAM (blue) are also stained.
