MMP3 antibody | knockout validation | Abcam ab52915

This is a knockout-validated antibody summary, based on the publication "MMP-3 Deficiency Alleviates Endotoxin-Induced Acute Inflammation in the Posterior Eye Segment", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit monoclonal IgG

Company: Abcam

Antibody: MMP3

Catalog number: ab52915

Summary: Rabbit monoclonal IgG against a synthetic peptide corresponding to human MMP3 (aa 450 to the C-terminus). Reacts with mouse, rat, and human. Suitable for western blot, immunohistochemistry (paraffin) and immunocytochemistry/immunofluorescence.

Validation Method

Western blot

Sample

Retina and retinal pigment epithelial (RPE) cells from WT and MMP-3−/− mice. Samples were homogenized in ice cold 100 µL lysis buffer (50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% Brij 35, and 1% Triton-X-100, pH 7.5), supplemented with protease inhibitor (Roche Applied Science, Mannheim, Germany).

Blocking agent

5% commercial blocking powder (ECL™ Blocking agent, GE Healthcare, Diegem, Belgium) for 2 h.

Primary incubation

1/200 dilution overnight at 4 °C.

Secondary incubation

Horseradish peroxidase (HRP)-labeled secondary antibodies.

Detection

Supersignal® West Dura Extended Duration Substrate solution (Thermo Scientific) and ChemiDoc MP Imager (Bio-Rad).
