WB: Cytosolic extracts prepared from the hearts of male MMP-2 knockout (KO) mice on a C57Bl/6J background (1, 21) and C57Bl/6J wild-type (WT) mice. IHC: heart sections from MMP-2 WT and KO mice. Tissues were fixed in 0.1% glutaraldehyde, 4% paraformaldehyde, and 4% sucrose in 0.1 M sodium phosphate buffer (pH 7.4) overnight at 4°C.
IHC: 1:10,000 overnight at 4°C.
WB: horseradish peroxidase-conjugated goat anti-rabbit IgG (CLCC42007, Cedarlane).
IHC: Colloidal gold (18 nm)-conjugated, donkey anti-mouse IgG secondary antibodies were used at a 1:10 dilution for 90 min at room temperature.
ECL Prime reagent (Amersham) and autoradiography film or scanned using a charge-coupled device-based imager (Carestream 4000 MM Pro Image Station).
