This is a knockout-validated antibody summary, based on the publication "Pancreatic cancer modeling using retrograde viral vector delivery and in vivo CRISPR/Cas9-mediated somatic genome editing", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Immunohistochemistry of tumors in Lenti-sgLkb1/Cre-infected KT;H11LSL-Cas9/+ mice lack Lkb1 protein. Positive control (pos) PDAC from an Ad-Cre-infected KP mouse and negative control (neg) tumor derived from a Lenti-sgLkb1/Cre-infected KT;Lkb1flox/flox mouse are shown. Bar, 50 μm. From [1].
Antibody information

Rabbit monoclonal IgG

Company: Cell Signaling

Antibody: Lkb1

Catalog number: 13031P

Summary: Rabbit monoclonal IgG against a recombinant protein specific to the carboxy terminus of human LKB1 protein. Reacts with human, mouse, and monkey by immunohistochemistry (paraffin).

Figure 2. Immunoblot of neoplastic cells from the pancreas of Lenti-sgLkb1/Cre-infected KT;H11LSL-Cas9/+ mice lack Lkb1 protein. KPT pancreatic cancer cells and a whole KT;H11+/+ pancreas were positive controls. Hsp90 shows equal loading. From [1].
Validation Method

Western blot | Immunohistochemistry

Sample

Neoplastic cells and pancreatic tumors from control and Lkb1-KO mouse. WB: cells were lysed with RIPA buffer (50 mM Tris pH 8, 150 mM NaCl, 0.1% SDS, 0.5% sodium deoxycholate, 1 mM NaVO4, and 1/100 diluted protease inhibitor cocktail (Sigma-Aldrich)). IHC: Tumors were fixed in 4% formalin and paraffin-embedded.

Secondary incubation

WB: HRP-conjugated anti-rabbit (Santa Cruz Biotechnology, sc-2004) antibodies.

Detection

IHC: Sections were developed with DAB.

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
