LZTR1 antibody | knockout validation | Santa Cruz sc-390166

This is a knockout-validated antibody summary, based on publications "RIT1 oncoproteins escape LZTR1-mediated proteolysis", as cited below [1], and "Impaired Proteolysis of Noncanonical RAS Proteins Drives Clonal Hematopoietic Transformation" for western blot knockout validation (figure 1e) [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1 (kappa light chain)

Company: Santa Cruz

Antibody: LZTR1

Catalog number: sc-390166

Summary: Mouse monoclonal IgG1 (kappa light chain) against a peptide mapping near the N-terminus of human LZTR1. Recommended for detection of LZTR1 of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence and ELISA. Also reactive with additional species, including equine, canine, bovine and porcine.

Validation Method

Western blot

Sample

Lztr1 WT (+/+), heterozygous (+/−), and KO (−/−) MEFs. Whole cell lysates were prepared using RIPA buffer (1% NP40, 150 mM NaCl, 0.5% deoxycholate, 0.1% SDS, 10

mM Tris-HCl pH=8).

Blocking agent

5% skimmed milk in TBST buffer for 1 hour.

Primary incubation

1:200 dilution overnight.

Secondary incubation

HRP-linked secondary antibodies.

Detection

Amersham ECL Western Blotting Detection Reagent.

Figure

Immunoblot of the indicated proteins in Lztr1 WT (+/+), heterozygous (+/−), and KO (−/−) MEFs. Each lane represents an independent cell line derived from a single embryo (two lines per genotype). Please see Figure 4a in the article [1].
