Sample

Bouin fixed samples of testes from wild-type and Ly6a -/- mice.

Blocking agent

5% rabbit serum in Coons buffer.

Primary incubation

Anti-LY6A/E overnight in blocking buffer at 1:50 dilution (fluorescence) or 1:200 dilution (DAB staining).

Secondary incubation

Fluorescent readout: readout: rabbit anti-rat IgG (BA4000, Vector Labs) followed by Texas red-conjugated goat anti-rabbit.

For staining: biotinylated rabbit anti-rat IgG followed by avidin-HRP conjugate.

Detection

For fluorescent readout: VECTAshield (Vector Labs) with Leitz DMIRB fluorescent microscope and Leica TCS4D confocal laser scanning microscope.

For staining: diaminobenzine.

See figure 1 in [1]. In the article, the authors do not show data for this antibody but state that the results were the same as for anti-LY6A/E (BD Biosciences 553333) which was also used in the paper and for which they present data confirming its specificity.
