Western blot

Sample

HeLa and LAMP2-deficient HeLa cells. Cells were lysed or homogenized in lysis buffer containing 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 5 mM EDTA, 1% Triton X-100 (Sigma-Aldrich, T8787) and protease inhibitors (Complete, EDTA-free; Roche Diagnostics, 11873580001) at 4°C.

Primary incubation

Overnight at 4°C

Secondary incubation

Horseradish peroxidase (HRP)-conjugated anti-rat IgG antibodies (Jackson ImmunoResearch, 112-035-003).

Detection

Using SuperSignal West Dura extended duration substrate (Thermo Scientific, 34075) and detected with a chemiluminescence imaging system (ImageQuant LAS 4000; Fujifilm).
