Validation Method

Immunocytochemistry

Sample

WT and Cre-mediated Kdm1a-deleted mouse embryos. Embryos were fixed in 4% paraformaldehyde, 0,2% sucrose, 0.04% Triton-X100 and 0.3% Tween20 in PBS for 15 min at 37°C.

Blocking agent

1% BSA, 0.05% Triton-X100 and 30 min in 1% BSA in 0.05% Triton-X100; 1 mg/ml polyvinyl pyrrolidone (PVP; Sigma).

Primary incubation

IF: 1:750 dilution in blocking buffer for ∼16 hr at 4°C.

Secondary incubation

1:500 dilution of Alexa568 goat anti-rabbit IgG/ Invitrogen A11036 for 2 hr at room temperature.

Detection

Inverted confocal microscope Zeiss (Germany) LSM700 with a Plan apo DICII (numerical aperture 1.4) 63x oil objective.
