Sample

Pancreata of wild-type and K8-deficient (K8−/−) mice. Pancreata were fixed in 4% buffered formaldehyde (Agar, Stansted, UK) for 24 h, embedded, cut into 4 µm thick sections and subjected to antigen retrieval by boiling in EDTA buffer.

Blocking agent

2% BSA.

Primary incubation

1∶50 dilution for 1 h at room temperature.

Secondary incubation

Fluorophore-conjugated antibodies.

Detection

confocal laser-scanning microscope (Zeiss LSM 710) equipped with Plan-Apochromat 63x and 40x/1.4 NA oil-immersion objective lenses, using the ZEN software.
