Sample

Skin from wild-type and Krt1−/−/Krt10−/− mice. WB: Total protein was extracted from E18.5 skin in sample buffer (125 mM Tris-HCl, pH 6.8, 10% glycerol, 1% SDS). IHC: Cryosections (5 μm) of neonatal and E18.5 skin were fixed for 10 minutes with pre-cooled acetone at −20°C.

Primary incubation

WB/IHC: 1:10 dilution.

Secondary incubation

WB: Horseradish-peroxidase-conjugated species-specific secondary antibodies (Dianova).

IHC: Species-specific Cy3-, Cy2-, Alexa-Fluor-594- or Alexa-Fluor-488-conjugated goat antibodies (Molecular Probes, Eugene, OR).

Detection

WB: Pierce Super Signal West Pico and Dura substrates (Thermo Scientific).

IHC: Axio Imager Z2 operated through Axiovision software (Zeiss, Hertfordshire, UK).
