Heart from control and Rbp2-KO mice. Tissues were thawed very briefly on ice and homogenized using TissueLyser II (Qiagen) in EBC lysis buffer (50 mM Tris-Cl pH 8.0 containing 0.5% vol/vol Nonidet P-40 and 250 mM NaCl) supplemented with a protease inhibitor mixture (Roche).

Blocking agent

5% nonfat milk in TBST (20 mM Tris-Cl pH 7.6 containing 150 mM NaCl and 0.1% Tween-20) for 1 h at room temperature.

Primary incubation

1:1,000 dilution in 5% BSA in TBST overnight at 4 °C.

Secondary incubation

At room temperature for 1 h.

Detection

Immobilon Western Chemiluminescent HRP Substrate (EMD Millipore) and developed using an ImageQuant LAS4000 chemiluminescence detector (GE Healthcare Life Sciences).
