Integrin β1 antibody | knockout validation | Millipore MAB2259Z

This is a knockout-validated antibody summary, based on the publication "Estrogen's Effects on Excitatory Synaptic Transmission Entail Integrin and TrkB Transactivation and Depend Upon β1-integrin function", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1

Company: Millipore

Antibody: Integrin β1

Catalog number: MAB2259Z

Summary: Mouse monoclonal IgG1 against human activated integrin β1. Reacts with human protein recognizing a cation- and ligand-induced binding site (LIBS), which has been used in some systems as a reporter of integrin functionality. Suitable for western blot, immunohistochemistry, immunoprecipitation, flow cytometry, blocking/activating experiments and cell culture.

Validation Method

Western blot

Sample

Hippocampus from control floxed and β1-cKO mice.

Primary incubation

1:400 dilution.

Secondary incubation

1:1,000 dilution AlexaFluor 488 anti-mouse IgG (Life Technologies, Carlsbad CA).

Detection

Epifluorescence image z-stacks were collected at 63X in 200 nm steps through 2 um from CA1b stratum radiatum (4-5 stacks/slice)

Figure

Deconvolved images of immunolabeling for PSD95 (green) and β1 integrin (red) in CA1 stratum radiatum show the dramatic reduction in punctate β1-immunoreactivity in a cKO mouse relative to a paired floxed-β1 WT; retention of some β1-ir is consistent with continued expression by glia and inhibitory neurons (bars = 3 μm). Please see Figure 4c in the article [1].
