Validation Method

Immmunocytochemistry.

Sample

Cells grown on coverslips were fixed in 4 % paraformaldehyde in phosphate buffered saline (PBS) for 10 minutes, and then permeabilized by incubation with 0.15 % Triton X-100 in PBS for 15 minutes.

Non-specific binding sites were blocked by incubating cells in a solution of 2 % BSA in PBS for 1 hour.

Primary incubation

1 hour at room temperature or overnight at 4 °C.

Detection

A Leica SP5 confocal microscope (Leica Microsystems, Wetzlar, Germany) equipped with a 63× oil immersion fluorescence objective.
