Huntingtin antibody | knockout validation | Abcam ab109115

This is a knockout-validated antibody summary, based on the publication "Ablation of huntingtin in adult neurons is nondeleterious but its depletion in young mice causes acute pancreatitis", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit monoclonal IgG

Company: Abcam

Antibody: Huntingtin

Catalog number: ab109115

Summary: Rabbit monoclonal IgG against a synthetic peptide corresponding to human Huntingtin aa 1-100. Reacts with mouse, rat, and human. Suitable for western blot, immunocytochemistry/immunofluorescence, immunohistochemistry (paraffin) and flow cytometry.

Validation Method

Western blot

Sample

Brain tissues from control and ubiquitous and neuronal Htt-KO mice. Protein samples were homogenized in the Nonidet P-40 buffer (50 mM Tris pH 7.4, 50 mM NaCl, 0.1% Triton X-100, 1% Nonidet P-40) or RIPA buffer (50 mM Tris, pH 8.0, 150 mM NaCl, 1 mM EDTA, 1 mM EGTA, pH 8.0, 0.1% SDS, 0.5% DOC, and 1% Triton X-100) with 1× protease inhibitor (Sigma, P8340).

Detection

SuperSignal West Femto Maximum Sensitivity Substrate (34095) or Amersham ECL Prime Western Blotting Detection Reagent (RPN2232).

Clone note

The same clone (EPR5526) is sold as Abcam ab109115; EMD Millipore MABN1105.
