This is a knockout-validated antibody summary, based on the publication "Endoplasmic reticulum-resident E3 ubiquitin ligase Hrd1 controls B-cell immunity through degradation of the death receptor CD95/Fas", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit monoclonal IgG

Company: Abcam

Antibody: Hrd1

Catalog number: ab170901

Clone: EP7459.

Summary: Rabbit monoclonal IgG against a synthetic peptide within human SYVN1 aa 150-250. Reacts with human, mouse and rat. Suitable for western blot, immunocytochemistry/immunofluorescence, flow cytometry and immunoprecipitation. Unsuitable for immunohistochemistry (paraffin).

Validation Method

Western blot

Sample

Primary B cells (B220+) isolated from bone marrow of WT and Hrd1 KO mice.

Blocking agent

4% milk dissolved in TBST.

Secondary incubation

Horseradish peroxidase-conjugated secondary antibodies.

Detection

Enhanced chemiluminescence detection system (ECL; GE Healthcare).

Figure

Please see Figure Suppl. Figure 1c in the article [1].

Notes

The article lists two different SYVN1/Hrd1 antibodies (Abcam ab170901 and Sigma SAB4200423). The figure 1c does not indicate which antibody was used. Since Sigma SAB420042 does not react with mouse, thus Abcam ab170901 is assumed. The antibody shows a nonspecific band.

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
