This is a knockout-validated antibody summary, based on the publications "mTOR promotes pituitary tumor development through activation of PTTG1", as cited below [1] and "Regulation of Intrinsic and Bystander T Follicular Helper Cell Differentiation and Autoimmunity by Tsc1" for western blot knockout validation (figure 1a) [2] and "Poly-L-arginine promotes asthma angiogenesis through induction of FGFBP1 in airway epithelial cells via activation of the mTORC1-STAT3 pathway" for western blot knockout validation (figure 3f) [3]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).
Rabbit monoclonal IgG
Company: Cell Signaling
Antibody: Hamartin/TSC1
Catalog number: 6935
Summary: Rabbit monoclonal IgG against a synthetic peptide corresponding to residues surrounding Val640 of human TSC1. Reacts with human, mouse and rat. Suitable for western blot and immunoprecipitation.
Western blot
Pituitary glands from control loxed and Ghrhr-Cre;Tsc1LoxP/LoxP mice and Tsc1−/− MEFs. Whole cells were lysed with lysis buffer (2% SDS, 10% glycerol, 10 mM Tris (pH 6.8) and 100 mM DTT).
HRP-labeled goat anti-rabbit antibody (Santa Cruz Biotech, sc-2004).
Chemiluminescence (NC15079) was from Thermo Scientific (Waltham, MA, USA).
Immunoblotting analysis of Tsc1 knockout in pituitary glands of Ghrhr-Cre; Tsc1LoxP/LoxP and Tsc1LoxP/LoxP mice and Tsc1−/− MEFs. β-actin served as a protein loading control. Please see Figure 2b and 5b in the article [1].
