Western blot

Wt and HSP70.1/3−/− MEFs. Cells were homogenized in sodium dodecyl sulfate (SDS) gel sample buffer (62.5 mM Tris, pH 6.8, 10% glycerol, 3% SDS), containing protease and phosphatase inhibitors.

5% nonfat milk in Tris-buffered saline with 0.05% Tween-20 (TBS-T).

1:3,000 dilution overnight.

Horseradish peroxidase (HRP)-linked secondary anti-mouse antibodies (Cell Signaling Technology Inc.) for 2 hr.

Enhanced chemiluminescence detection kit (PerkinElmer Life Sciences).
