Validation Method

Immunohistochemistry

Sample

Coronal brain slices of 3-month control and Hdac3 cKO male mice. Mice were perfused with 10% formaldehyde under deep anesthesia and brains were post-fixed overnight in 10% formaldehyde. Brains were sectioned at 40 μm using a vibratome (Leica).

Blocking agent

PBS containing 0.3% Triton X-100 and 10% normal donkey serum at room temperature (20–22 °C) for 1 h.

Primary incubation

1:200 dilution in blocking buffer overnight at 4 °C.

Secondary incubation

Alexa Fluor 488, Alexa Fluor 568, and Alexa Fluor 647 antibodies diluted 1:500 in PBS and incubated at 20–22 °C for 90 min.

Detection

Images were acquired using an LSM 710 Zeiss confocal microscope.
