Sample

Aorta samples from control HDACfl/fl mice or vascular smooth muscle-specific SM22-cre;HDAC1fl/fl mice. Tissues were homogenized in lysis buffer ((50 mM Tris (pH 8.0), 150 mM NaCl, 1 mM EDTA, 1% NP-40, 1 mM DTT, 1 mM phenylmethylsulfonyl fluoride, 1 mM Na3VO4 and 1 μg ml−1 each of leupeptin, pepstatin and aprotinin).

Blocking agent

5% skim milk in 1 × TBST.

Primary incubation

1:1,000 dilution overnight at 4 °C.

Secondary incubation

Horseradish peroxidase-linked secondary antibodies for 1 h at room temperature.

Detection

Enhanced chemiluminescence using a Fuji LAS-3000 system (Fujifilm Life Science, Tokyo, Japan).
