Antibody information

Rabbit monoclonal IgG

Company: Abcam

Antibody: Granulin

Catalog number: ab187070

Summary: Rabbit monoclonal antibody against a recombinant fragment of mouse Granulin (PGRN). Reacts with mouse. Suitable for western blot, immunohistochemistry (paraffin), immunocytochemistry/immunofluorescence, immunoprecipitation and flow cytometry (intra).

Validation Method

Western blot | Immunohistochemistry

Sample

WT and PGRN KO mouse placental tissues.

Blocking agent

WB: 5% skimmed milk/PBS for 1 h at room temperature.

IHC: 10% H2O2 for 10 min at room temperature, followed by 5% goat serum (cat. no. SP-9001/SP-9002; SPlink Detection kits, Biotin-Streptavidin HRP Detection Systems; OriGene Technologies, Inc.) at 37°C for 30 min.

Primary incubation

WB: 1:1,000 dilution overnight at 4°C.

IHC: 1:200 dilution overnight at 4°C.

Secondary incubation

WB: 1:5,000 dilution horseradish peroxidase (HRP)-conjugated goat anti-rabbit IgG secondary antibody (cat. no. SH-0031/0011; Beijing Dingguo Changsheng Biotechnology Co., Ltd.) for 1 h at room temperature.

IHC: non-diluted biotinylated goat anti-rabbit IgG polymer working solution (cat. no. SP-9001/SP-9002; SPlink Detection kits, Biotin-Streptavidin HRP Detection Systems; OriGene Technologies, Inc.) at 37°C for 30 min.

Detection

WB: Immobilon Western Horseradish Peroxidase (Merck KGaA), enhanced chemiluminescence western blotting reagents (cat. no. RPN2109; Cytiva) and a Bio-Rad ChemiDoc MP Imaging system.

IHC: horseradish peroxidase-conjugated streptavidin for 30 min at 37°C, and diaminobenzidine staining. Images were captured using an Olympus IX81 upright light microscope.
