Synaptoneurosomes from hippocampi and a part of adjacent cerebral cortex of WT and Fmr1 KO mice. Tissues were lysed in 10 mM HEPES, pH 7.4, 200 mM NaCl, 30 mM EDTA, 0.5% Triton X-100 with protease inhibitor cocktail cOmplete (Roche, Indianapolis, IN) and 80 U/ml RiboLock RNase Inhibitor (Thermo Scientific).

Blocking agent

5% BSA and 3% normal goat serum (NGS) in PBST for 1 h at RT.

Primary incubation

1:1,000 dilution overnight at 4 °C with gentle shaking.

Secondary incubation

1:10,000 dilution peroxidase-labeled secondary antibodies (Vector Laboratories, Burlingame, CA) in 5% BSA in PBST for 1 h at RT.

Detection

Enhanced chemiluminescence using Amersham ECL Prime Western Blotting Detection Reagent (GE Healthcare) and Amersham Imager 600 (GE Healthcare). S
