Validation Method

Western blot

Sample

WT and CRISPR-induced Flad1-KO Huh7.5.1 cells. Cell pellets were lysed using Laemmli SDS sample buffer containing 5% beta-mercaptoethanol.

Blocking agent

5% non-fat milk, 0.1% Tween-20 PBS .

Primary incubation

1:250 dilution in blocking buffer overnight at 4 C rotating.

Secondary incubation

1:5,000 dilution HRP-conjugated secondary anti-mouse antibodies (Genetex) for 1 hr at room temperature.

Detection

West Pico and Extended Duration peroxide solutions (Pierce) and visualized on film.

Notes

RPS25 (Abcam, 102940), MAGT1 (Proteintech, 17430-1-AP), RPN1 (gift from Martin Ivessa), STT3B (Sigma, HPA036646), ELAVL1 (Santa Cruz, sc-5261), RFK (Sigma, SAB1409492) and SRRD (Sigma, HPA002945) are also KO validated (WB), although some of them are polyclonals.
