Validation Method

Immunohistochemistry

Sample

White adipose tissue cells from wild-type and ERB-2 -/- mice.

Cells were treated with 4% paraformaldehyde overnight, washed with PBS and then incubated overnight with PBS containing 5% Donkey serum and 0.5% Triton X-100.

Blocking agent

PBS containing 5% Donkey serum and 0.5% Triton X-100.

Primary incubation

Overnight in blocking buffer. Antibody used at 1:200 dilution.

Secondary incubation

Secondary antibody (Alexa Fluor conjugated, Life Technologies) incubated in blocking buffer for 2 hours at room temperature.

Detection

Leica TCS Sp8 confocal microscope.
