E4BP4 antibody | knockout validation | Santa Cruz sc-74414

This is a knockout-validated antibody summary, based on the publication "E4BP4 is an insulin-induced stabilizer of nuclear SREBP-1c and promotes SREBP-1c-mediated lipogenesis", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Notes

The antibody described could also be sc-28203, a discontinued rabbit polyclonal antibody from Santa Cruz. Validation data for polyclonal antibodies can not be applied to future or past lots/batches. Antibody information

Mouse monoclonal IgG2a (kappa light chain)

Company: Santa Cruz

Antibody: E4BP4

Catalog number: sc-74414

Summary: Mouse monoclonal IgG2a (kappa light chain) against amino acids 163-462 mapping at the C-terminus of human E4BP4. Recommended for detection of human E4BP4 by western blot, immunoprecipitation, immunofluorescence and ELISA.

Validation Method

Western blot

Sample

Control and E4bp4 knockout primary mouse hepatocytes. The modified RIPA buffer (50 mM Tris (pH 8.0), 150 mM NaCl, 2 mM EDTA, 0.5% NP-40, 0.05% SDS, 0.25% sodium deoxycholate, and protease inhibitor cocktail) was used to lyse cells and extract protein.

Detection

Immunoreactive bands were detected using homemade ECL reagents: 1.25 mM Luminol, 0.2 mM p-coumaric acid, 0.1 M Tris-HCl (pH 8.5).
