DJ-1 antibody | knockout validation | Cell Signaling 5933

This is a knockout-validated antibody summary, based on the publications "Efficient bi-allelic gene knockout and site-specific knock-in mediated by TALENs in pigs", as cited below [1], and "DJ-1 suppresses ferroptosis through preserving the activity of S-adenosyl homocysteine hydrolase" for western blot knockout validation (figure 2a, s1g) [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

DJ-1 antibody | knockout validation | Cell Signaling 5933 figure 1
Figure 1. Western blot results in six tissues of wild-type and KO piglets. +/− represents mono-allelic KO piglet (8-day-old), −/− represents bi-allelic KO piglet (2-day-old), and +/+ represents wild type pigs (10-day-old, female). β-tubulin and β-actin served as housekeeping gene controls. From [1].
Antibody information

Rabbit monoclonal IgG

Company: Cell Signaling

Antibody: DJ-1

Catalog number: 5933

Summary: Rabbit monoclonal IgG against a synthetic peptide corresponding to residues surrounding Lys148 of human DJ-1 protein. Reacts with human, mouse, rat, hamster, and monkey. Suitable for western blot, immunoprecipitation and immunofluorescence/immunocytochemistry.

Validation Method

Western blot

Sample

Tissues of wild-type and DJ-1KO piglets. Samples were lysed in a buffer containing 50 mM HEPES, pH 7.4, 150 mM NaCl, 100 mM NaF, 0.05% SDS, 1% Nonidet P-40, 1 mM phenylmethylsulfonyl fluoride, 2 g/ml aprotinin and leupeptin, 2 mM benzamidine, 1 mM sodium vanadate, 1 mM EGTA and 0.1% deoxycholate for 15 min on ice.

Primary incubation

1:1,000 dilution.
