Validation Method

Immunohistochemistry

Sample

WT and Dgki-/- mouse DRG tissue.

Blocking agent

10% normal donkey serum (S30, EMD-Millipore, Burlington, Massachusetts) in TBST (NDS/TBST) for 1 h at room temperature (approximately 23°C).

Primary incubation

1:300 dilution in NDS/TBST overnight at room temperature.

Secondary incubation

Blocked again with NDS/TBST for 30 min, followed by 1:500 goat anti-mouse IgG2a-Alexa 488 (A21131, Thermo Fisher Scientific) in NDS/TBST for 6 h at room temperature.

Detection

Imaging was performed on a Zeiss LSM 710 confocal laser scanning microscope.
