Sample

Heart from control (f/f), cardiac-specific constitutive (cKO) and inducible (iKO) DGAT1 KO mice. Whole heart proteins were extracted from flash frozen hearts using 10–20 mg of wet weight tissue using RIPA buffer containing 150 mM NaCl, 50 mM Tris-HCl, 1% Triton X-100, 0.1% SDS, 0.1% sodium deoxycholate and 1 mM EDTA, pH 7.4.

Primary incubation

Overnight at 4 °C.

Secondary incubation

Anti-mouse HRP-conjugated secondary antibody (R&D Systems).

Detection

Enhanced chemiluminescence (ECL) detection solution (Supersignal Westpico Chemiluminescent Substrate, Thermo, #34080) and exposed to X-ray film (HyBlot CL, Denville Scientific, Inc., NJ, US).
