Sample

WB: gastric tissues from wild-type, TFF1 KO, DARPP-32 KO, and TFF1 KO/DARPP-32 KO mice. Cells were lysed with RIPA buffer complemented with protease and phosphatase inhibitors that were purchased from Santa Cruz.

IHC: gastric tissues from wild-type, TFF1 KO, DARPP-32 KO, and TFF1 KO/DARPP-32 KO mice. Formalin-fixed paraffin-embedded.

IF: gastric organoids from wild-type, TFF1 KO, DARPP-32 KO and TFF1 KO/DARPP-32 KO mice.

Primary incubation

IF: 1:400 dilution overnight at 4°C.

IHC: primary antibody overnight at 4°C.

Secondary incubation

IF: 1:800 dilution Alexa Fluor goat anti-mouse IgG (Invitrogen) at room temperature for 2h.

IHC: 1 h incubation at room temperature.

Detection

IF: inverted laser-scanning LSM880-Airyscan (Zeiss, Thornwood, NY) confocal microscope.

IHC: horseradish peroxidase activity was identified using a Histostain Plus kit (Invitrogen), following supplier’s instructions.
