Summary: Mouse monoclonal to P Glycoprotein. specific for the MDR1 isoform of P Glycoprotein which is only found in the plasma membrane. It does not cross-react with MDR3. It has been shown to cross-react with Pyruvate Carboxylase, an abundant mitochondrial enzyme (130 kDa), on both immunoblots and immunohistochemical tissue sections. Weak homogeneous, cytoplasmic, or granular patterns of reactivity may represent staining of the PC cross-reactive epitope rather than positive staining for P glycoprotein. Reacts with mouse, rat, and human and has been used for western blot, ELISA, immunocytochemistry, immunohistochemistry (paraffin), immunoprecipitation and flow cytometry.

Validation Method

IHC, immunohistochemistry

Sample

Female P-glycoprotein (P-gp) knockout (KO) CF-1 mice. Tissue was fixed with 10% formalin for 24 hours and paraffin sections processed by heat-induced epitope retrieval.

Blocking agent

Slides were blocked with 5% normal goat serum/phosphate-buffered saline (PBS) for 30 minutes.

Primary incubation

Overnight incubation with rabbit anti-P-gp (1:4000) or with Abcam mouse anti-P-gp antibody JSB-1 (1:20).

Detection

Detected with the streptavidin-biotin immunoperoxidase method and diaminobenzidine substrate for visualization.

Notes

Both rabbit P-gp antibody made by the researchers and mouse (JSB-1, ab3366) antibody show similar staining patterns and both are considered validated.
