Validation Method

Immunohistochemistry

Sample

Cochlea from wildtype and CtBP2 KO mice.

Blocking agent

4% (w/v) BSA/PBS at room temperature for 2 hr.

Primary incubation

1:400 dilution in blocking buffer overnight at 4˚C.

Secondary incubation

1:600 dilution anti-mouse secondary antibodies (Alexa Fluor 488, 546 or 555, 647) in 4% [w/v] BSA/PBS) for 1 hr at room temperature.

Detection

Confocal microscope (LSM700 or LSM880, Zeiss) using 1.4 NA oil immersion objectives for confocal imaging, and using 1.2 NA water immersion objective for Airyscan imaging.
