Western blot | Immunohistochemistry

Coronal sections and lysates of the hippocampus from 6-week-old WT or Cpne6 KO mice. For WB, hippocampi were dissected on ice and homogenized in lysis buffer (50 mM Tris-HCl pH 7.5, 5 mM EDTA, 150 mM NaCl, 1% NP-40 and 0.5% sodium deoxycholate, including proteases and phosphatase inhibitor cocktails (Roche)) by a glass/Teflon homogenizer. For IHC, brains were embedded in paraffin and 5-μm-thick sections were cut on a microtome.

IHC: 5% BSA and 0.2% Triton X-100 in PBS for 30 min at room temperature (RT).

WB: 1:1,000 dilution.

IHC: 1:200 dilution in blocking solution overnight at 4 °C.

IHC: for 1 h at RT.

IHC: Epifluorescence microscopic pictures were obtained with a Leica DM5000B fluorescence microscope and analySIS software (Soft Imaging System).
