This is a knockout-validated antibody summary, based on the publication "N-cadherin haploinsufficiency affects cardiac gap junctions and arrhythmic susceptibility", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit polyclonal

Company: Zymed, now part of ThermoFisher

Antibody: connexin 43

Catalog number: 71-0700

Summary: rabbit polyclonal antibody raised against a peptide corresponding to a segment of the 3rd cytoplasmic domain (C-terminal portion) of rat connexin 43. It detects rat, mouse and human connexin 43. Supplier recommended for elisa, immunofluorescence, immunohistochemistry and Western blotting.

Detects total connexin 43.

Validation Method

Western blotting

Immunohistochemistry

Sample

Western blot: homogenized hearts from connexin 43 -/+ heterozygous mice

Immunohistochemistry: hearts from connexin 43 -/+ heterozygous mice. Samples were prepared with formalin in PBS, pH 7.4 then embedded in paraffin wax. Epitopes were unmasked by treating with 1 x Antigen Unmasking Solution (Vector Labs).

Blocking agent

Western blot: not stated.

Immunofluorescence: 5% non-fat milk in PBS for 30 minutes.

Primary incubation

Western blot: not stated.

Immunohistochemistry: 1:100 overnight at 40C in blocking solution.

Secondary incubation

Western blot: alkaline phosphatase-conjugated secondary antibody.

Immunohistochemistry: Alexa Fluor 555-conjugated goat anti-rabbit IgG (Molecular Probes) for 1 hour.

Detection

Western blot: chemifluorescence (ECF, Amersham Biosciences) with densitometry using Image-Quant software (Molecular Dynamics).

Immunohistochemistry: confocal microscopy with Zeiss LMS510 Meta confocal microscope.

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
