This is a knockout-validated antibody summary, based on the publication "Recruitment and activation of the ATM kinase in the absence of DNA-damage sensors", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Antibody: Ku70

Catalog number: 4588

Cell Signaling rabbit monoclonal Ku70 antibody 4588 was validated in knockout MEFs figure 1
Figure 1. Immunoblots of Ku70 levels in Ku70+/+ and Ku70-/- MEFs, from Figure S1 [1].
Summary

Rabbit mAb produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val294 of mouse Ku70 protein. It has been shown to react with human, mouse, rat, and monkey and it is predicted to react to hamster, bovine and pig, based on 100% sequence homology. It has been used in western blot.

Validation Method

Western blot.

Sample

Cell extracts of mouse embryonic fibroblasts (Ku70 knockout, and control cells) were prepared in RIPA buffer (150mM NaCl; 1% NP-40; 0.5% C24H39O4Na; 0.1% SDS; 50mM Tris-HCl, pH 8.0) or Laemmli buffer (4% SDS; 20% glycerol; 120 mM Tris-HCl, pH 6.8), resolved by SDS-PAGE and transferred using standard procedures.

Secondary incubation

IRDye-conjugated goat anti-rabbit (Li-Cor Biosciences).
