Cdk1FLOX and and Cdk1NULL MEFs. Cells and tissues were lysed in EBN buffer: 80 mM β-glycerophosphate (pH 7.3), 20 mM EGTA, 15 mM MgCl2, 150 mM NaCl, 0.5% Nonidet P-40, 1 mM DTT, and protease inhibitors (20 μg/mL each of leupeptin, chymostatin, and pepstatin; EI8, EI6, and EI10; Chemicon).
Tris-buffered saline with 0.1% Tween 20 and 5% nonfat dry milk (1706404; Bio-Rad).
Overnight at 4 °C

Protein was still detected in the KO liver (Authors say the signal was from other cell types). The figure is not a good example of KO validation. Fig 4b is after a IP with Cdk1-deficient MEFs, and looks better.
