Cells were lysed using Cell Lysis Buffer (#9803; Cell Signaling Technology) containing: 20 mM Tris-HCl (pH 7.5), 150 mM NaCl, 1 mM Na2EDTA, 1 mM EGTA, 1 % Triton, 2.5 mM sodium pyrophosphate, 1 mM β- glycerophosphate, 1 mM Na3VO4 (phosphatase inhibitor) and 1 µg/ml leupeptin (protease inhibitor), supplemented with 5 mM NaF (phosphatase inhibitor), 10 µg/ml aprotinin (inhibitor of trypsin and related proteases), 10 µg/ml Pefabloc SC (proteinase K inhibitor) and 1 mM DTT, a reducing agent.

Blocking agent

TBS containing 5 % bovine serum albumin and 0.1 % Tween 20 for 1 h at room temperature.

Primary incubation

1:1,000 dilution overnight at 4 °C.

Secondary incubation

Horseradish peroxidase-conjugated goat anti rabbit IgG polyclonal antibodies (111-035-003; Jackson ImmunoResearch Laboratories).

Detection

SuperSignal West Pico chemiluminescence substrate (Perbio, Bonn, Germany).
