Validation Method

Immunocytochemistry

Sample

Cby1+/+ or Cby1−/− MEFs.

Blocking agent

PBS containing 3% bovine serum albumin (Sigma-Aldrich), 0.1% Triton X-100, and 0.02% sodium azide (PBS-BT).

Primary incubation

1:100 dilution in PBS-BT for 1–3 h at room temperature or overnight at 4°C.

Secondary incubation

Alexa Fluor dye–conjugated secondary antibodies (Invitrogen) were diluted in PBS-BT 1:500 and incubated at room temperature for 45 min.

Detection

images were acquired using an Axiovert 200M microscope (Carl Zeiss, Jena, Germany) with Plan-Neofluar 100×/1.3 numerical aperture (NA) and PlanApoChromat 63×/1.4 NA objectives and a cooled, charge-coupled device (CCD) camera (Orca ER; Hamamatsu Photonics, Hamamatsu, Japan).
