Sample

WT and CTSD-KO ARPE-19 cells.

Blocking agent

5% nonfat dry milk in Tris-buffered saline (TBS) (50 mM Tris, 150 mM NaCl, pH = 7.6) containing 0.1% Tween-20 (Sigma-Aldrich) (TBS-T) for 1 hour.

Secondary incubation

1:5,000 horseradish peroxidase-conjugated secondary antibody in blocking buffer for 2 hours at room temperature.

Detection

chemiluminescence ECL Prime Western Blotting Substrate (GE Healthcare, Chicago, IL) and imaged using a Chemidoc XRS (Bio-Rad).
