Blocking agent

5 % milk in Tris Buffered Saline Tween solution (TBST; 20 mM Tris, 0.5 M NaCl, and 0.05 % Tween 20, pH 7.4) for 1 h

Primary incubation

1:1000 diluted in Super Block T20 (Thermo Scientific). at 4 C overnight

Secondary incubation

goat-anti-rabbit-horseradish peroxidase (1:10,000; Thermo Scientific, Cat # 32460) diluted in TBST for 1 h

Detection

visualized in a FluorChem 8900 imager (Alpha Innotech, San Leandro, CA) using ECL detection reagent Super Signal West Femto Maximum Sensitivity Substrate (Thermo Scientific).
