Sample

Bone marrow-derived dendritic cells (BMDC) from WT and Casp1/11-/- mice. Whole cell detergent lysates were prepared by addition of 56 ul of RIPA lysis buffer (0.5% sodium deoxycholate, 0.1% SDS, 1% IgePal CA630 in PBS, pH 7.4, plus protease inhibitor mixture). Extracellular medium samples were concentrated by trichloroacetic acid precipitation/acetone washing.

Primary incubation

1 ug/ml dilution.

Secondary incubation

0.13 ug/ml dilution HRP-conjugated secondary antibodies.

Detection

Chemiluminescent images of the developed blots were detected, stored, and quantified using a FluorChemE processor (Cell Biosciences).
